Inhibition of Nicotinic Receptor-Mediated Catecholamine Secretion by Some Medicinal Herbs

Abstract

Effect of the water extract from several oriental medicinal herbs and theirs groups which are related to inhibition stress, on catecholamine secretion was investigated in bovine adrenal chromaffin cells. The water extract of Sosiho, Tang among several herbs groups tested inhibited [^3H]norepinephrine([^3H]NE) secretion induced by a nicotinic acetylcholine receptor (nAChR) agonist DMPP with a half maximal inhibitory concentration(IC5o) of ¡6§¶/ml. Each elemental herbs of Sosiho, Tang also were investigated in other consentration( 0.1 §·/ml, 1 §·/ml). Panax ginseng C.A.¡¤Meyer Pinellia ternata BREIT¡¤Zingiber officinale Roscoe in 1 §·/ml inhibited [^3H]norepinephrine([^3H]NE) secretion.
However, cytosolic calcium([Ca^2+]i) induced by DMPP was difficult to investigate in Fluorescence measurement because that each elemental herbs of Sosiho, Tang had yellowish colour. Though Machilus thunbergii Sieb. is not a elemental herb of Sosiho, Tang, inhibited exactly [^3H]norepinephrine([^3H]NE) secretion in 1 §·/ml consentration and inhibited cytosolic calcium([Ca^2+]i) induced by DMPP in dependency on consentration.
